Middle would be better but opposite side to spare wheel if there is one there. Just make sure the weight doesn't cause any problems including putting the van's ATM over weight.
Me, I try not to put too much weight on the rear of the van at all. I only have the spare wheel and bumper.

A couple of things to beware of putting a large (heavy) item on the back of the van.
1 It may upset van stability under tow by lowering tow-ball weight and increasing pendulum moment.
2 It must not obscure number plate.

Dougwe is quite right regarding weight at the rear end of your van being a wobbly issue.
I would be inclined to advise that the offside(nearest the centre line of the road) would be better than near the door if your van is a rear door model.
Ensure the exhaust iss pointed away from the van if possible to reduce the noise inside.

Any weight on the end of a van can seriously destabilise it. I would advise against it.
Much better to put it in the back of the tug or on the A frame.

Noise reduction is all about density plus overall thickness plus isolation
2 layers of plywood with acoustic glue between them and then pyramid foam on the inside .
Allow enough room for cooling air and exhaust air .
Isolation mount the box to draw bar or rear bumper .
The above is for noise reduction .
Placing gene in anything less just changes the tone .
